Experimental Production of Antibody to Procaine

Abstract
Procaine was emulsified with complete Freund adjuvant and used for sensitization of ten¢¥ albino rabbits about weighing 2500 gm. The rabbits were injected intradermally, intramuscularly and sensitized again by smear on the scarified dorsal skin simultaneously for five consecutive days.
Four weeks after the last sensitization, the rabbits were bled partially, and antibodies to procaine in sera were tested by precipitation, complement fixation, hemagglutination, modified hemolysis, and Prausnitz-Kustner tests. One day after the partial bleeding, skin tests were performed by intradermal in;ection of procaine, tetracaine, PABA, and penicillin. The sensitized rabbits were also subjected to anaphylactic shock for procaine. The results obtained were summarized as follows.
1. Ring precipitation and agar-gel diffusion tests, neither precipitin rings nor bands were detected in sera of sensitized rabbits.
2. The sera of sensitized rabbits showed negative complement fixation reactions, but seven out of ten sensitized rabbits developed serum antibodies, which were detected by hemagglutination and modified hemolytic tests.
3. In sensitized rabbits, the intradermal skin tests and anaphylactic shock to procaine run parallel to certain extent.
4. The positive results of the blocking test by Prausnitz-Kustner technique suggested the production of specific procaine antibodies in sera of sensitized rabbits.
5. The positive results of active intradermal tests by injecting both of PABA and tetracaine as antigens suggest that the aromatic ring of PABA may be the major antigenic determinant group responsible for the cross reaction to the procaine hypersensitivity in rabbits.
